Using the Nexus 7: The Kindle Fire's got something to fear

1 day

Google's Nexus 7 is a strange device. At 7?inches, it's smaller than an iPad ??and a breadbox, for that matter ??and with its impressive spec list, it's a bit more appealing than the similarly low-priced Kindle Fire. But how does it hold up in person?

Surprisingly well.

While?I cringe when I cradle the Kindle Fire's clunky body,?the Nexus 7 feels perfectly balanced and secure in my hand. It's a solid device?? the unit I'm using survived several awkward drops off a table's edge ? and it looks sleek.

On the inside, the Nexus 7 has a quad-core processor, a twelve-core GPU and 1 GB of RAM. Between that hardware and Project Butter???under-the-hood improvements which keep?Jelly Bean running particularly?smoothly?? the tablet's pretty darn speedy. (For more about Jelly Bean, check out?our rundown of its best features.)

When Google announced the Nexus 7, it pushed its content consumption features ? especially?a widget which collects books, magazines, TV shows, movies and music. It's an appealing tool and, like most things on the Nexus 7, it is consistently smooth and brisk.

I've only used the Nexus 7 for a couple of days now, but I already dread returning home and reaching for a Kindle Fire. I know that it'll feel slow, clunky, and unsatisfying in comparison.

Rumor: Amazon prepping new 7- and 10-inch Kindle tablets

1 hr.

A larger version of Amazon's popular Kindle Fire tablet is being readied for release, an anonymous source tells BGR. The company is also planning to revise the original with an improved metal body and upgraded specs.

The Kindle Fire is far and away the single most popular Android-based tablet, though Amazon replaced most of the user-facing part of Google's OS with its own interface and apps. Although the company's policy is not to state sales numbers, it's clear that millions have been sold, and the gamble of competing with Apple's iPad?? by offering a smaller, cheaper tablet???paid off.

But there is clearly demand for larger tablets as well, and rumors have persisted that Amazon was working on a 10-inch tablet the entire time, but deemed it too risky to launch both. Now that the Fire has proven successful, it's less risky to put a more expensive option out there, and BGR's source claims this is just what they are doing.

The 10-inch device is said to be equipped with a quad-core processor, which would make it a more powerful media playback machine than the current Fire, which some now?regard as slightly underpowered. It would also have a front-facing camera, presumably for video chat purposes.

Both the larger and the smaller devices will have high-quality metal builds, an improvement on the Fire's slightly plasticky feel. Both will also have HDMI-out ports for plugging the device into your TV or monitor, and neither one will have any buttons on the front.

No timing or pricing data was suggested by the anonymous source, but it would be reasonable to assume that the smaller one will remain around $200, if not a little less, and the larger one will go for $100 to?$150 more, carefully avoiding competing directly with the year-old?iPad 2, which still sells in an 8GB version at $399. Expect a large announcement sometime fairly soon, though, at which time we may also expect the glowing?Kindle leaked out in April.

21% of iOS users say they wouldn't leave Apple at any price

By Neil Hughes

Published: 03:24 PM EST (12:24 PM PST) iPhone and iPad users are so loyal to Apple and unlikely to switch to another platform that one analyst has pegged their total collective value at nearly $295 billion.

Bill Shope with Goldman Sachs said in a note to investors on Friday that he believes the average single iOS customer is worth $1,053. That assumes the average selling price of an iOS device for the June quarter is $535 with 5 percent erosion, gross margin is 45 percent, and annual defections are 5 percent.

"This implies a cumulative iOS customer value of nearly $295 billion on today's installed base, without any consideration for the content, services or peripherals streams, or for the platform's growth potential," Shope wrote.

At a value of $295 billion, iOS customers represent about 55 percent of Apple's current market cap. That's before including the company's $110 billion cash balance, an iOS install base that continues to grow at a tremendous pace, or the value of the company's Mac and iPod businesses.

Goldman Sachs recently conducted a consumer survey of over 1,000 respondents, in an effort to better understand the costs that customers face when they switch platforms.

The survey found that 21 percent of respondents said no discount, regardless of the amount, would make it worthwhile to leave Apple's platform. Among those who said they would consider switching, more than half of those said they would need a discount greater than 30 percent.

Among those polled, 71 percent said they are "highly likely" to choose an Apple device for their next tablet or smartphone purchase. Another 23 percent are "likely" to stick with Apple's ecosystem, while only 1 percent were "unlikely" to stick with Apple.

Switching costs for customers only grow when they own more than one device in the Apple ecosystem, and the new iCloud service adds yet another layer to the switching costs, Shope said. Among those polled by Goldman Sachs, 30 percent said they are paying for extra storage capacity or iTunes Match with iCloud.

"Considering that apple has disclosed that it has 120 million iCloud users as of its earnings release on April 24, this is quickly becoming yet another high-margin revenue stream for the company," Shope said. "More important, however, this also suggests that a large portion of Apple's installed base is adding yet another source of explicit switching costs."

In all, he estimates that the switching cost per iOS user ranges between $122 and $301. That implies the average user would need a discount of 49 percent to switch to a competing platform.

Lochte, Phelps set for memorable 200 free final

OMAHA, Neb. (AP) ? Ryan Lochte and Michael Phelps went stroke for stroke into the wall in the 200-meter freestyle. Lochte's hand got there first, with Phelps touching second.

Imagine what Wednesday night's final will be like.

The world's top two swimmers met up in the same semifinal Tuesday night, with Lochte winning in 1 minute, 46.25 seconds. Phelps was just behind at 1:47.27, setting up another memorable confrontation in the final.

"It's going to be a tough race tomorrow," Phelps said. "It's probably going to be another close one like that."

Phelps will be looking to win for the first time at the U.S. Olympic trials. He got beat by Lochte in the 400 individual medley on Monday, then got edged out again in the semis.

Phelps is the ultimate competitor, hating to lose even the smallest contest. But he knows what's really at stake in Omaha.

"It's about getting spots on the team, that's the only thing," he said. "The most important thing for me is to try and secure a spot, and from here I have 3 to 4 weeks to prepare myself and get some extra training in there. I think it's going to be a lot better than what it is here."

Lochte agreed nothing truly matters until the final.

"We're great racers, we just want to win," he said. "We definitely kicked it in gear the last 50, me and him. We know tomorrow night is going to be a lot faster."

There promises to be plenty of speed in the women's 100 backstroke final.

Seventeen-year-old Missy Franklin was the top qualifier in the semifinals. Her time of 59.06 seconds was less than a second off Gemma Spofforth's world record. Teenagers claimed the next three fastest times, as well: 18-year-old Rachel Bootsma, 17-year-old Olivia Smoliga, and 18-year-old Elizabeth Pelton.

"I love where I'm at right now," said Franklin, the breakout star at last year's world championships and one of the top American hopes for London. "I feel strong, I feel powerful. It's so awesome to feel this way and I'm having a blast. This is what I came here to do."

Natalie Coughlin finds herself in the unexpected role of underdog in her signature event. The two-time defending Olympic champion barely qualified for the final with the seventh-fastest time of 1:00.63. That race came after Coughlin finished seventh in the 100 fly final.

"I was hoping to have a better 100 fly than I did, but you pay for it when you go out too hard like I did," Coughlin said. "After that, I just refocused on the 100 backstroke, knowing all that matters is I get a lane for tomorrow night. We'll see what happens. That's all I can do right now."

Coughlin won't have another event before the 100 back final, like she did Tuesday. She scratched the 200 individual medley, so if she doesn't qualify for London in the backstroke, her only realistic chance left to make the team will be the 100 freestyle.

"Having the morning off to rest and recover and go into that 100 back as fresh as possible will be really important for me," she said.

Olympic champion Rebecca Soni used her patented finishing kick to qualify fastest for the 100 breaststroke final in 1:05.88, more than a second ahead of world-record holder Jessica Hardy, who was second in 1:06.88.

"I always hold on to that last few yards as my strength," Soni said. "I'm always waiting to kick it into gear. I don't want to do it too early, but it felt good coming home, and I'm looking forward to having that same speed closing tomorrow."

Hardy won the 100 breast at trials four years ago only to lose her spot on the Olympic team because of a failed doping test. She served a one-year ban after an arbitration panel agreed with her contention that a tainted nutritional supplement was to blame for her positive test.

She battled anger and depression during the fight to clear her name.

"This is the first time I felt like I can actually have a happy ending," she said. "I can relax and finally have fun and feel grateful."

Matt Grevers is the favorite in the men's 100 backstroke final on Wednesday, going in with the top semifinal time of 53.10. Lochte had the second-quickest time in prelims, but he dropped out of the semis to focus on his next final against Phelps.

Brendan Hansen capped his comeback with a victory in the 100 breast, earning a spot in his third straight Olympics.

He won in 59.68.

Hansen is relishing having another shot at Japanese star Kosuke Kitajima, who swept both breast events at the last two Olympics and is in Omaha for the trials so he could keep up his training under American coach Dave Salo.

"It's cool that he got to see me swim," Hansen said. "Somewhere deep down, he's got to know I'm the only guy to beat him the last 10 years. That might carry some weight."

Hansen retired from the sport after the Beijing Games, having been disappointed in two straight Olympics. He took silver and bronze in the two breast events at Athens and was shut out of an individual medal in China.

"I don't know if my body language has shown it or not, but this is exactly where I want to be," he said.

"In 2008, I didn't really want to be here anymore, I was kind of worn down by the whole idea. Now I'm just having so much fun with it and I feel like I'm running on borrowed time. I'm just really proud of myself for saying that I was going to come back and then actually come back and be back."

Hansen will be joined on the team by Eric Shanteau, who specializes in the 200 breast but claimed the second spot in the 100 in 1:00.15.

Four years ago, Shanteau beat out Hansen for an individual spot on the team shortly after being diagnosed with testicular cancer. He put off treatment until after the games and has been healthy ever since.

"I've been back and forth with that event for so long," Shanteau said. "It's nice having it all come together when it counts the most."

Dana Vollmer, a gold medalist as a teenager in 2004, missed out on the team four years ago while battling injuries and health problems, but will go to London after her win in the 100 butterfly.

Vollmer was more than a half-second under world-record pace at the turn, but she faded on the return lap. But she had built such a commanding lead that she was a full body length ahead when she touched in 56.50.

Claire Donahue claimed the second spot for London in 57.57.

Also claiming a spot on the Olympic team was Allison Schmitt, who got off to a huge lead in the 400 free and held on to win in 4:02.84. Chloe Sutton earned the second spot in 4:04.18.

Quest Software gets higher takeover bid of $2.32B

(AP) ? Quest Software Inc. said Monday that it received a higher buyout offer worth about $2.32 billion from an unnamed buyer, ramping up the battle for the technology services provider.

The most recent offer of $27.50 per share follows a string of increasing bids that began after the company announced in March that it was being bought by investment firm Insight Venture Partners for $23 per share. At that time, the Aliso Viejo, Calif., company said the deal valued it at about $2 billion.

The March bid was a 19 percent premium to the company's closing stock price the day before the deal was announced. The $27.50 per share offer announced Monday would represent a 42 percent premium to Quest's ending price of $19.40 from March 8.

Shares of Quest, which helps companies manage databases and provides other corporate IT services, rose $1.48, or 5.6 percent, to close at $27.70 Monday. The stock earlier hit a 17-month high. That suggests investors think there may be yet another, higher offer coming.

Quest said Monday that its board has decided that the most recent $27.50 per share bid is superior to prior offers and that it has informed Insight and Vector Capital, which was added to the buyout group this month. Insight and Vector have three days to match the offer.

While Quest did not say Monday who the other bidder was, reports published last month said that Dell Inc. was in discussions with Quest about a potential transaction. Quest declined to comment further on who was behind the bid announced Monday. Dell also declined comment.

Earlier this month, an undisclosed party offered to buy the company for $25.50 per share, or about $2.15 billion. Last week, Insight and Vector topped that with a bid of $25.75 per share.

In the event that shareholders who previously supported the deal with Insight do not support the most recent offer, the company will grant the unnamed bidder the right to buy new shares of the company's stock equal to a 19.9 percent stake, or pay it a break-up fee under certain circumstances.

Quest's CEO Vinny Smith had agreed to support Insight's offer in March, which would allow him to roll his 34 percent stake in Quest into the private company and remain CEO.

Quest declined to say Monday if there were other shareholders involved.

Smith became CEO in February.

HBT: Ump blows double-switch, costs Cards

The Cardinals beat the Marlins in extras last night, but umpire Bob Davidson made it all the harder when he screwed up a double-switch made by Mike Matheny which cost the Cards Allen Craig.

The short version: when Matheny put reliever Victor Marte into the game, he signaled ?five? with his hands, meaning that he was taking over David Freese?s position in the lineup, with Freese being the third baseman or the five-position, defensively (he was batting seventh). After a batter had batted, however, Marlins manager Ozzie Guillen came out and said told Davidson that Matheny meant the five-hole in the lineup, which had been occupied by Allen Craig.

Davidson inexplicably agreed with Guillen regarding what Matheny meant, said that the pitcher was supposed to be batting fifth now and that Allen Craig had to leave the game. This later led to the Cardinals having to use a pitcher to pinch hit in extra innings. Which worked out ? he got an RBI ? but still.

Later the on-field mics picked up Davidson telling Ozzie that he ?f****d up,? and that Matheny had, in fact, told him that he wanted the pitcher in the seven-hole. Too late, of course.

Just another day in Mr. Davidson?s neighborhood.

Sony NSZGS7 Google TV review

If at first you don't succeed, try, try again and that's exactly what Google has done with Google TV. With the second generation of Google's software hitting older hardware and some TVs, it only makes sense that Sony would revise its hardware and fill the void left when Logitech decided to leave the party. The $199 NSZ-GS7 from Sony might be the perfect little box for those hoping to internet-enable their TV without actually replacing the TV itself. How well it does at that is exactly what you'll find if you click through.

BREAKING: Aaron Walker Victory in Maryland Emergency Hearing ...

Posted on | June 25, 2012 | 64 Comments

FROM AN UNDISCLOSED LOCATION
At a hearing this morning in Rockville, Maryland, a Montgomery County Circuit Court granted an emergency motion on behalf of attorney/blogger Aaron Walker. The effect of the ruling will permit Walker to resume writing and speaking about convicted terrorist bomber Brett Kimberln.

Judge Nelson W. Rupp Jr. presided over today?s hearing, with attorney Reginald W. Bours III representing Walker, who did not attend the hearing at Judicial Center in Rockville. In a brief telephone interview, Walker said that Kimberlin filed a counter-motion that did not address the First Amendment legal issues at stake, but rather ?made a bunch of new accusations? against Walker.

It is important to note that Judge Rupp?s ruling only temporarily permits Walker to speak and write about Kimberlin, pending a July 5 hearing in Maryland state appeals court. Walker is appealing a May 29 ruling by Montgomery County District Court Judge C.J. Vaughey that granted a peace order sought by Kimberlin against Walker.

Walker said he plans to file a report on today?s hearing at his blog, Allergic to Bull. A Virginia attorney, Walker has become the center of national attention since May 17, when he published a 28,000-word blog post entitled, ?How Brett Kimberlin Tried to Frame Me for a Crime (And How You Can Help!).? Walker?s post highlighted Kimberlin?s?long-running efforts harass and intimidate bloggers who had written about Kimberlin?s criminal history.

Kimberlin is director of a 501(c) non-profit, the Justice Through Music Project, which has collected more than $1.8 million in contributions since 2005. Kimberlin also co-founded another tax-exempt activist group, Velvet Revolution, that has made headlines (and raised money) by making accusations of wrongdoing?against a variety of public figures, including GOP strategist Karl Rove, U.S. Chamber of Commerce President Tom Donohue, Supreme Court Justice Clarence Thomas, and the late Andrew Breitbart.

In addition to his appeal in Maryland state courts, Walker has filed for an emergency preliminary injunction in federal court.

As the legal blog Popehat notes, UCLA Law Professor (and popular blogger)?Eugene Volokh has agreed to provide pro bono assistance to Walker?s legal defense team. University of Tennessee Law Professor Glenn Harlan Reynolds,?whose blog Instapundit is one of the most popular center-right sites,?has also shown strong?support for Walker.

Patrick ?Patterico? Frey also promises to provide more?commentary about the Walker-Kimberlin case. Walker, writing under the pseudonym ?Aaron Worthing,? was formerly a co-blogger at Patterico.com, a popular conservative site.

Award-winning conservative blogger Ace of Spades comments on today?s Maryland ruling. Ace was one of the targets of an escalating pattern of harassment against bloggers that evidently began last year, during the Anthony Weiner cybersex scandal.

Democrat ?social media? consultant Neal Rauhauser has described Kimberlin as a ?client.? Rauhauser evidently joined forces with Kimberlin?s Velvet Revolution group last summer?and is suspected of?involvement with a Web site, originally aimed at ?exposing??Breitbart, which published items?indicating?an ongoing effort to determine the offline identity of Ace. The anti-Breitbart site has been repeatedly published personal information (including home addresses) of bloggers who wrote about Brett Kimberlin?s criminal past.

Rauhauser became obsessed with a conspiracy theory explanation of the so-called WeinerGate scandal, which resulted in a formerly popular Democratic congressman from New York resigning in disgrace. Rauhauser has boasted of his connections to the criminal computer-hacking ring known as ?Anonymous.?

Both Mike Stack, who played a key role in exposing Weiner, and Patterico ??blamed as a prime suspect in Rauhauser?s conspiracy theory of the WeinerGate scandal ? were targeted?by a harassment?tactic known as?SWATting, a dangerous criminal hoax. In a May 25 blog post, Patterico named Rauhauser, Kimberlin and former Raw Story editor Ron Brynaert among those he suspects of complicity in the bogus 911 call that resulted in a police raid on Patterico?s Los Angeles home.

Aaron Walker describes his own situation: ?An Important Victory For Freedom of Expression,? which includes a link to?Judge Rupp?s order. Aaron says to expect further updates.

(Note: This article was originally posted at 10:46 a.m. ET as a single paragraph, and subsequently updated seven times over the course of three hours as new information was added. I have now removed the ?update? notices for ease of reading.)

UPDATE 10 p.m. ET: Aaron Walker was SWATted tonight.

> Insurance > Insurance ? Why Should You Go For It?

Insurance is a form of risk management. It can be defined as the transfer of a risk to an insurance company in exchange for a premium paid by the insured at regular intervals. Insurance is useful both for the insurer and the insured. The insurer earns profit by investing the money of the insured and getting returns on it. The insured, on the other hand, has the security of the sum assured which he will earn at the end of the insurance term.

In United Kingdom, you get insurance products like motor insurance, home insurance, pet insurance, life insurance, travel insurance, health insurance and business insurance etc. motor insurance covers both the driver of the car and the car itself. Health insurance covers medical bills incurred due to illness or accidents. Travel insurance covers certain losses such as medical expenses, loss or theft of personal belongings while traveling, traveling delay etc. pet insurance will cover your pet?s medical expenses, loss or death. Home insurance covers your house and in some cases its contents. Damages arising due to natural calamities like earthquake, floods etc are excluded. You are also not supposed to get the assured sum when the damage caused was an ?act of god? or a result of war. Business insurance covers losses that you may incur in your business.

The type of risk involved to the risk determines how much will be the premium. For example, if you have bought a home insurance and your house is in good shape and situated near a fire station, the insurance company will charge you lower premiums. Risk factor therefore determines the premium amount.

UK ONLINE MARKET is a guide to hundreds of insurance companies who offer you attractive cash back offers on your purchasing their insurance products. We have merchants for every type of insurance available: motor insurance, home insurance, life insurance, travel insurance, health insurance, business insurance, and pet insurance. Foam Party | Shoptalk | Gambit New Orleans News and Entertainment

The summer heat is upon us, and the right insulation can make the difference between a poorly or properly cooled home. At Green Apple Foam (25 Papworth Ave., Metairie; 408 N. Columbia St., Covington, 258-2464; www.greenapplefoam.com) Ed and Jo Ann Lozes specialize in spray foam insulation, a technique widely used in commercial properties. Only recently has it been used in homes.

??The benefits of spray foam insulation can be seen on homeowners' electric bills and taxes. "Spray foam is the highest-performing air barrier, and it carries the highest (energy) ratings per square inch," Ed says. "It reduces the HVAC [heating, ventilation and air conditioning] load by as much as 35 percent. This saves on installation costs on new construction projects ? and saves as much as 50 percent on monthly utility bills for both new construction and retrofits."

??Tax incentives give homeowners additional motivation to make their homes more energy efficient. The Louisiana Home Energy Rebate Option (HERO) program, Energy Smart and other programs offer homeowners a chance to get money back after a spray foam installation. Most homeowners can receive a federal tax credit of $500.

??Benefits also include stopping wall condensation, mold and mildew, and improving indoor air quality. "A typical attic, on a hot summer day, with no shade, it's about 135-138 degrees," Ed says. "Spray an adequate amount of foam in there, it should be ... about 85-88 degrees. Because you're dropping that heat load by such a percentage, the air-conditioning unit is not working nearly as hard, and it's delivering much cooler air."

??Ed and Jo Ann both have extensive experience in the construction and home contracting business, and each brings their own special expertise to the company. Jo Ann uses her degrees in journalism and business administration to give each client special care. Ed began his own painting company when he was 21, and expanded into building custom homes. "After [Hurricane Katrina], we were building in the Gulf Coast area and there wasn't anyone spraying foam insulation the way we thought it should be done," he says.

??For the Lozes, owning a business in their hometown is a satisfying feeling. "We're very proud of our reputation in the New Orleans area," Lozes says. "We've been in the home improvement industry for over 30 years, and we've built our reputation on customer satisfaction. A job is never complete until the homeowner or contractor is completely satisfied."

??The company's name has an eco-friendly ring, which reflects the way Ed and Jo Ann feel about their work. "Green apples carry a healthy, clean and wholesome connotation," Ed says. "This is how we want people to feel about our company. We're honest contractors with a wholesome reputation, and we leave every project broom-swept clean."

The spreadsheet is easy to determine. Either worksheet if they are to be completed. Many parents enjoy the limits set by the spreadsheet, but it is best for your child?

Ask yourself the following question:

Is my child working on a task for the trash?
Does my child have a better understanding of the issue by completing this task?
Is my child happy, the task at hand?
Does my child looking for a way to share tasks with others? Are they proud of their work?
There is a better way. Let me show you note in the book arts. Not to remove. You have to be more creative! Phew! All you need is a rich collection of books, articles, or a healthy supply of textbooks and supplies interesting, and your child will do basically the rest.

Adding Creativity Learning Using Notebook

Adding Creativity Learning Using Notebook

Well, maybe not so simple, but almost. Your child knows when something is just a time filler. I?m guilty of this form of training. If you already homeschooling for a long time you will realize that this is true. When I give my children a sheet of notebook paper and tell them to write something, they more than likely to scribble on a blank page to stare for some time at the edges.

Notice Reservation template is a very different answer. There is something magical about a neat and creative piece of paper with the organized space is managed for a variety of information. There are thousands of designs from the booking immediately available knowledge. I have a template of my hand to my kids today. Even a simple box for illustration and explanation of a few lines out of the way my kids a task to challenge the change. Well, if they read a story to get to or from an event in history, or scientific experiments were carried out to have beaten for them to the point of them as the most attractive control, then write and illustrate a few good words to to describe what they take.

The first thing I noticed is that the templates are managed in order to improve the most time to produce. The second thing I noticed is that children can not wait to show off their work. I started experimenting with the style and presentation templates. I started their pages in plastic protectors and notebook page. A funny thing happened. The more time I get to showcase their work, applied the more children the next time when the task is to ?book? to attack them head on. If the worksheet is easy to give them a second look when finished.

(AP) ? A giant Pacific octopus that's the star attraction at an Ohio zoo's new reef exhibit has chosen her name by pulling shrimp from a labeled ball.

A list of 2,200 suggestions submitted to the Akron Zoo in a public naming contest was narrowed to three options Monday for the 20-pound, 4-foot-long octopus, and she chose Cora. It's a shortened version of coral, which is a popular octopus habitat and the theme of the exhibit.

The Akron Beacon Journal (http://bit.ly/LvJ404 ) reports the octopus made her decision by selecting the ball labeled Cora, pulling out shrimp, and then holding up the winning name with a tentacle as photographers captured the moment.

The most-submitted name suggestion had been Octavia. The other option, Scarlet, referred to the creature's reddish skin.

Most commonly mutated gene in cancer may have a role in stroke

ScienceDaily (June 22, 2012) ? The gene p53 is the most commonly mutated gene in cancer. p53 is dubbed the "guardian of the genome" because it blocks cells with damaged DNA from propagating and eventually becoming cancerous. However, new research led by Ute M. Moll, M.D., Professor of Pathology at Stony Brook University School of Medicine, and colleagues, uncovers a novel role for p53 beyond cancer in the development of ischemic stroke. The research team identified an unexpected critical function of p53 in activating necrosis, an irreversible form of tissue death, triggered during oxidative stress and ischemia.

The findings are detailed online in Cell.

Ischemia-associated oxidative damage leads to irreversible necrosis which is a major cause of catastrophic tissue loss. Elucidating its signaling mechanism is of paramount importance. p53 is a central cellular stress sensor that responds to multiple insults including oxidative stress and is known to orchestrate apoptotic and autophagic types of cell death. However, it was previously unknown whether p53 can also activate oxidative stress-induced necrosis, a regulated form of cell death that depends on the mitochondrial permeability transition pore (PTP) pore.

"We identified an unexpected and critical function of p53 in activating necrosis: In response to oxidative stress in normal healthy cells, p53 accumulates in the mitochondrial matrix and triggers the opening of the PTP pore at the inner mitochondrial membrane, leading to collapse of the electrochemical gradient and cell necrosis," explains Dr. Moll. "p53 acts via physical interaction with the critical PTP regulator Cyclophylin D (CypD). This p53 action occurs in cultured cells and in ischemic stroke in mice. "

Of note, they found in their model that when the destructive p53-CypD complex is blocked from forming by using Cyclosporine-A type inhibitors, the brain tissue is strongly protected from necrosis and stroke is prevented.

"The findings fundamentally expand our understanding of p53-mediated cell death networks," says Dr. Moll. "The data also suggest that acute temporary blockade of the destructive p53-CypD complex with clinically well-tolerated Cyclosporine A-type inhibitors may lead to a therapeutic strategy to limit the extent of an ischemic stroke in patients."

"p53 is one of the most important genes in cancer and by far the most studied," says Yusuf A. Hannun, M.D., Director of the Stony Brook University Cancer Center, Vice Dean for Cancer Medicine, and the Joel Kenny Professor of Medicine at Stony Brook. "Therefore, this discovery by Dr. Moll and her colleagues in defining the mechanism of a new p53 function and its importance in necrotic injury and stoke is truly spectacular."

Dr. Moll has studied p53 for 20 years in her Stony Brook laboratory. Her research has led to numerous discoveries about the function of p53 and two related genes. For example, previous to this latest finding regarding p53 and stroke, Dr. Moll identified that p73, a cousin to p53, steps in as a tumor suppressor gene when p53 is lost and can stabilize the genome. She found that p73 plays a major developmental role in maintaining the neural stem cell pool during brain formation and adult learning. Her work also helped to identify that another p53 cousin, called p63, has a critical surveillance function in the male germ line and likely contributed to the evolution of humans and great apes, enabling their long reproductive periods.

'Brave' hits bull's-eye at the box office

By Pamela McClintock, The Hollywood Reporter

Pixar

"Brave" earned $66.7 million on its opening weekend.

Fears that a female heroine would slow down "Brave" proved unfounded as the animated tentpole opened to a whopping $66.7 million at the domestic box office -- the fifth best debut of all time for a Pixar title.

"Brave,"?continuing Pixar's unblemished record of opening all of its movies to No. 1, also scored the second highest June opening for an animated pic after Pixar's "Toy Story 3" ($110.3 million). Overseas, the Pixar and Disney title debuted to $13.5 million in 10 markets.

The 3D event pic -- receiving an A CinemaScore in North America -- marks Pixar's 13th film and is the first movie in the company's history to feature a female lead. "Brave" did skew female (57 percent), but got plenty of male attention.

"You have to draw men and boys as well to see this number," Disney executive president of worldwide distribution Dave Hollis said. "The themes in the movie -- bravery, fighting for your fate -- transcend gender."

Hollis credited Pixar/Disney animation chief John Lasseter, producer Katherine Sarafian and directors Mark Andrews and Brenda Chapman for delivering an "enveloping experience" that drew both families (66 percent) and adults.

One troubling statistic: 3D revenues only made up 34 percent of "Brave's"?opening gross, furthering worries that families find the upcharge for a 3D ticket too expensive.

Still, family product ruled the box office all the way around. DreamWorks Animation and Paramount holdover "Madagascar 3: Europe's Most Wanted" fell to No. 2 in its third weekend with an estimated $20.2 million for a domestic cume of roughly?$157.6 million ("Madagascar 3"?opened to $60.1 million).?

Animation also ruled overseas, where "Madagascar 3" stayed at No. 1 for the third weekend in a row, grossing $30.1 million from 44 markets to race past the $200 million mark. The pic has now earned $208.4 million internationally for an impressive worldwide total of $366 million.

Managing only a third-place domestic finish was 20th Century Fox's 3D genre epic "Abraham Lincoln: Vampire Hunter." The R-rated film, playing like a horror title, grossed a soft $16.5 million in its opening. Fox had predicted a debut in the $15 million range -- considering there are no big stars in the film --? but box office observers believed it could get to $20 million.

Directed by Timur Bekmambetov and featuring Tim Burton in the producer's seat, "Abraham Lincoln" received a C+ CinemaScore. The pic's cast is led by Benjamin Walker.

"Abraham Lincoln,"?costing $68 million to produce, features the storied U.S. president as a vampire hunter and is based on the novel by?Seth Grahame-Smith, who also wrote the adapted screenplay. The film is an important test for the "mash-up" genre, with Lionsgate queued up to make the film adaptation of Grahame-Smith's book "Pride and Prejudice and Zombies."

Fox believes "Abraham Lincoln,"?which skewed male, will have good legs.

"Audiences will continue to seek out Timur's daring and brilliant vision of 'Abraham Lincoln,' " Fox's incoming president of domestic distribution Chris Aronson said.

The news wasn't good for Steve Carell-Keira Knightley indie pic "Seeking a Friend for the End of the World," which debuted to $3.8 million from 1,625 locations. The Focus Features title, directed and written by Lorene Scafaria, came in No. 10, just ahead of fellow Focus pic "Moonrise Kingdom," which grossed a pleasing $3.4 million from only 395 theaters for a pleasing cume of $11.6 million.

"It's disappointing. The right people turned up to see "Seeking a Friend," but they didn't come in volume," Focus president of distribution Jack Foley said.

At the specialty box office, Woody Allen's new entry "To Rome With Love" got off to a strong start, grossing $379,371 from five theaters for a sizeable location average of $75,874 -- easily the best of the weekend. Sony Pictures Classics, which distributed Allen's box office hit "Midnight in Paris," is once again handling domestic distribution duties for the filmmaker.

Elsewhere, New Line and Warner Bros.' troubled musical "Rock of Ages" fell to No. 6 in its second weekend, grossing $8 million for a 10-day domestic cume of $28.8 million. Adam Sandler's comedy "That's My Boy," likewise troubled, fell to No. 7 in its second outing, grossing $7.9 million for a 10-day total of $28.2 million.
